Do I have enough staggered joints in my tile installation, or should there be more for proper alignment?

Normally we try to align tapered to tapered joints and butt joints together, but what you’re doing will work fine.
I’m not a big fan of using backer board on floors, but if you thinset it to a clean and stable subfloor, use Durock screws every 8” in both directions, and thinset/tape all the seams, you should be okay.
Your layout looks good to me. I’m not a fan of using cement board on wood floors. I prefer an uncoupling membrane. As mentioned before, make sure the subfloor is clean and stable. Also, check the thickness of the subfloor – the cement board requires 5/8” to 3/4” thick depending on floor joists spacing. Use a good thinset bed to lay the boards, mesh tape/mud the seams, and use plenty of screws. You should be good.
